- Sam Raimi signed on to direct the sequel immediately after completing the first Spider-Man film.
- Sony hired writers from the TV series Smallville because director Raimi, producer Laura Ziskin, and star Tobey Maguire all enjoyed the show.
- The film was originally titled 'The Amazing Spider-Man' but this served only as a placeholder, with alternative titles like 'Spider-Man: No More' and 'Spider-Man Unmasked' proposed instead.
- Michael Chabon's script draft featured Doc Ock becoming infatuated with Mary Jane Watson, with his mechanical limbs designed to use endorphins to counteract pain from being attached to his body.
- Sam Raimi drew thematic inspiration from Superman II when developing how Peter Parker would grapple with giving up his responsibilities.
- Tobey Maguire was nearly replaced by Jake Gyllenhaal after a pre-existing back condition was aggravated during the filming of Seabiscuit, but Maguire's girlfriend's father Ronald Meyer, who headed Universal Studios, helped him regain the role.
- Alfred Molina was cast as Doctor Octopus after Sam Raimi was impressed by his performance in Frida and felt his large physical size matched the comic book character.
